Structural Biology

Our lab aims to understand biological processes by looking at the structures of macromolecular complexes; to that end our predominant tool and expertise is in X-ray crystallography (MX) - actually a team integrated to the lab is at the forefront of developing methods for X-ray crystallography - but we also use Small Angle X-ray Scattering (SAXS), Nuclear Magnetic Resonance (NMR) and Electron Microscopy (EM). We complement our structural data with an array of biochemical methods and biophysical techniques, including Surface Plasmon Resonance (SPR), Isothermal Titration Calorimetry (ITC) and Multiple Angle Laser Light Scattering (MALLS).

Our main interests include DNA replication licensing and how that is regulated throughout the cell cycle, and the role of Polo-like kinases and their partners in cell cycle. We also have an active interest in the structure of Autotaxin and JBP1 on proteins which are exciting drug targets. More scientific data are available in the NKI research reports.

Computational macromolecular crystallography
ARP/wARP (in collaboration with the group of V. Lamzin at EMBL Hamburg), is a software suite that aims to automate building of macromolecular models to the electron density maps that are available after every X-ray crystallography experiment. A combination of empirical knowledge of protein structure, pattern recognition, linear combinatorics and image processing algorithms together with real and reciprocal space crystallographic refinement techniques are exploited for that purpose.

Future research plans, which are focused around the 'Structural genomics' initiatives, include the development of algorithms to facilitate model building at low-resolution maps, completion of partially known structures and design of better protocols for the end-users.
